Sequencing

 Techniques

When examining 800 genotypes per day, it is essential that one develop a set of techniques that is both efficient and accurate. The sequencing team utilizes Promega Read-it© technology to perform single nucleotide polymorphism (SNP) typing and to purify DNA samples.




Machines

The ABI 3700 (left) is used to perform sequence analysis and fragment analysis as well as providing data in several different formats (top). The ABI 3700 is capable of running four 384 well plates over a twenty four hour period or four 96 well plates over a twelve hour period achieving read lengths of 700 - 800 base pairs per sample, depending on the primer. The team also takes advantage of a Biomek FX in conjunction with an LJL Luminometer to organize, view, and interpret genotypic information.

 

Future

We continue to explore new means of high throughput sequencing. Additionally, we are pursuing innovative genotyping methods, including fluorescent and SNaPshot genotyping.
