Microarray

  • Provides rapid and cost-effective methods of identifying gene expression and genetic variation
  • Gene expression profiling: allows one to screen large numbers of genes to determine whether or not they are active under various conditions
  • Important for diagnosis and treatment of diseases
  • Provides ability to study pathways related to specific functions
  • Ability for quantitative analysis using ratios of expression levels for individual genes
  • Being used in a number of laboratory applications for a variety of projects

The Microarray Process

  • Select clones, replicate, and perform PCR
  • Coat slides w/ Poly-L-Lysine to adhere PCR product onto slide
  • Stamp clones onto slides using an arrayer (right)
  • RNA is extracted from tissue sample
  • RNA is reverse transcribed to cDNA
  • cDNA samples are tagged with fluorescent labels (ex: Cy3 and Cy5) and are applied to the array
  • Single stranded cDNA from sample bind (hybridize) to the complementary strand of cDNA stamped on slide
